Title:Highly Fluorescent, Photostable, and Ultrasmall Silicon Drug Nanocarriers for Long-Term Tumor Cell Tracking and In Vivo Cancer Therapy

Abstract:In this communication, we introduce the first example of a small-sized (4 nm), fluorescent and photostable nanocarrier based on SiNPs with strong fluorescence and robust photostability. The fluorescent SiNPs nanocarriers (SiNP-DOX) are prepared from SiNPs via a simple one-step synthesis, followed by loading with doxorubicin (DOX, a commercial chemotherapy drug for cancer treatment) molecules. Not that the fluorescent SiNPs nanocarriers are produced without requiring additional doping with fluorescent species. Significantly,the photostable SiNPs provide the first demonstration of fluorescent drug nanocarriers-based long-term live cell tracking. Furthermore,we show the fluorescent SiNP nanocarriers are superbly suitable for in vitro and in vivo cancer therapy. Typically, malignant tumor cells (e.g., Hela and MCF-7 cells) are almost completely killed by the fluorescent nanocarriers during 48-h incubation. Moreover, tumor-bearing mice treated by SiNP nanocarriers survive over 20 d without any observable tumor growth.
